Twin Siblings\’ Rare Growth Disorder Holds Clues to Preventing Cancer


In Piñas, a village nestled amid the Andes, two sisters of 40—María Luísa Romero and María del Cisne—face a lifelong battle with Laron syndrome. The disorder, a rare genetic mutation that stops them from growing taller than 3.9 feet, has paradoxically made them the subjects of a potentially life‑saving study.

Despite their short stature, the sisters say that having neighbors who share their condition makes coping easier. Yet research led by endocrinologist Dr. Jaime Guevara suggests more than just community support—immunity. In a 22‑year study comparing around 100 Laron patients to 1,600 normal‑height relatives, no cases of diabetes and only one case of non‑fatal cancer were recorded in the former group, while 5% of normal‑height participants developed diabetes and 17% faced cancer.


The key appears to lie in insulin‑like growth factor 1 (IGF‑1), a hormone produced in the liver that helps cells grow and divide. In Laron syndrome, the body cannot use growth hormone to stimulate IGF‑1 production, resulting in lower IGF‑1 levels. Lower IGF‑1 levels may suppress the cancer cells’ ability to evade apoptosis, the programmed cell death that normally keeps unchecked growth in check. Dr. Guevara hopes that if this mechanism can be replicated—through drug or diet— it might offer a safe way to lower cancer risk for people without Laron syndrome.


Beyond cancer, Laron syndrome's lack of IGF‑1 offers a natural perspective on aging and metabolic disorders. While the twins themselves had never experienced the growth spurts of childhood—missed by the age window for the drug Increlex, the first treatment that can stimulate growth in those aged 2–18—they report thriving in adulthood with a robust family life. Each has a child who did not inherit the syndrome, and all four share a common affection for their community’s traditions.


The promise of translating these findings to wider populations is tempered by practicality. Increlex cost can exceed $800 per bottle, necessitating several bottles per month for a child, making it financially prohibitive for most families. Nonetheless, for the twins—to accept their size as a part of who they are—studies keep promising a future where untreated Laron syndrome may still guide us toward the next breakthrough in cancer prevention.
